초록
The abnormal accumulation of Tau protein in the brain begins in the medial temporal lobe, including the entorhinal cortex and hippocampus, and gradually spreads to other cortical areas, showing a pattern of progression that is strongly correlated with memory impairment. Therefore, understanding the mechanisms underlying the initiation of Tau pathology and establishing techniques to control the spread of Tau pathology are urgently needed for the development of more realistic treatment strategies. In this study, a cell model capable of monitoring Tau protein aggregation was established, and a highthroughput screening platform was established using an automated imaging system with phosphorylated tau-specific antibodies and HCS (High Content Screening) equipment for efficient sample screening. Using the high-content screening system in the Tau protein aggregation neuronal cell mode. We performed screening using an FDA-approved drug library. About 20 drugs reduced tau protein aggregation by more than 40%. In the future, we will validate the therapeutic efficacy of those using in vivo mouse model of tauophathy.
